光合膜蛋白结构生物学
沈建仁
博士,课题组长,研究员;
1961年11月出生。1982年,浙江农业大学环境保护系,学士;1986年,日本东京农工大学环境保护系,硕士;1990年,日本东京大学生物系,博士。1990年至2003年,先后任日本理化学研究所博士后研究员、研究员、高级研究员。2003年,任日本国立冈山大学理学院生物系教授。2003年至2008年被日本理化学研究所聘为兼职研究员。2007年至2012年被日本兵库县立大学聘为特邀教授。2002年至2006年兼任日本科学技术振兴机构先驱研究项目研究员, 2011年至2012年被聘为日本科学技术振兴机构先驱研究领域顾问。2012年, 入选国家“千人计划”。2013至今,中国科学院植物研究所,研究员。

光合作用是藻类和高等植物利用太阳光能把水和二氧化碳转换成有机物,同时产生氧气的复杂过程。光合作用产生的有机物不仅给人类提供了赖以生存的食物,也是我们现在利用的各种化石能源的来源。光合作用的另一个产物氧气是大气中几乎所有氧气的来源,为包括人类在内的需氧生物提供了生存不可缺少的氧气,为现代人类的生存和繁荣提供了必要的基础。光合作用能量吸收、传递和转化过程中涉及4个重要的膜蛋白复合物。光系统II (Photosystem II,PSII) 是光能吸收、传递和利用中的第一个膜蛋白复合物,它利用光能来催化水裂解反应,产生质子、电子和氧气。其中的质子和电子为二氧化碳的固定提供了能量和还原剂, 而氧气则是我们放氧生物的生存必不可少的。因此,可以说PSII是一个极为重要的蛋白复合物。
最原始的放氧光合生物蓝藻的PSII由20个蛋白亚基组成, 其中17个是膜蛋白,3个是结合在膜表面的亲水性蛋白,总分子量达350 kDa。在原子水平上阐明PSII的结构和功能,由此来解明光合生物利用可见光分解水的原理,将不仅为阐明光合作用的机理提供重要的基础,也为人工模拟光合作用,利用太阳能制氢提供重要的理论依据,因此对开发清洁能源和改善环境有重要的意义。另外, 由于光系统II容易受到光的破坏,限制了光合作用效率,它的结构与功能的研究对于抑制光破坏,提高光合作用效率,增加粮食产量也有重要的应用价值。
鉴于PSII水裂裂和放氧反应在自然界演化和人类文明进程中的重要性,我们的主要研究方向为:光系统II的结构和功能, 膜蛋白高分辨率晶体的制备和其结构解析。
